Abstract: Tangguh (Pettiveria alliacea) is one of the medicinal plants used for cancer, hepatitis and diabetes. The increase use of medicinal plants for herb therapy causes in increase demand of raw materials. Somatic embryogenesis for mass production of callus derived from leaves was conducted. Leaves from three-months old seedling were cut into 1 x 1 cm pieces and used as explants. Leaves were sterilized by ethanol and bayclin then rinsed with sterile water twice. Explants were cultured on MS medium supplemented with sugar 30 g/l, agar 8 g/l and vitamin of group B. The experiment was conducted in three stages, callus initiation, induction of embryogenic callus and germination of embryogenic callus. To induce production MS medium supplemented with 2,4-D (0.1, 0.2 and 0.3 mg/l) was used. The best callus were subcultured to the same medium supplemented with 2,4-D 0,1 mg/l to obtain embryogenic callus. Embryogenic callus were subcultured into MS medium for maturation by using cytokinin BA (0.1, 0.3 and 0.5 mg/l), kinetin (1, 2 and 3 mg/l) or zeatin (0.5, 1.0 and 1.5 mg/l) to obtain somatic embryo and/or shoots. The observation was made on visual of callus, percentage of embryogenic callus and embryoid and number of adventitive shoots. The experiment was designed as Complete Randomized (CRD) with ten replicates. The results showed that the treatment wIT 2,4-d 0.1 mg/l was the best for callus initiation producing white, compact and friabel callus. Subcultured of callus to the same medium supported the development of the embryogenic with white and compact callus. The treatment with kinetin of 1 mg/l was the best for maturation of embryogenic callus to form shoots. Tangguh (Pettiveria alliacea) merupakan salah satu tanaman obat diantaranya untuk penyakit kanker, hepatitis dan diabetes melitus. Dengan meningkatnya penggunaan obat sebagai herb theraphy maka kebutuhan akan bahan baku juga meningkat. Penelitian mengenai embriogenesis somatik dari eksplan daun tanaman tangguh telah dilakukan. Bahan tanaman yang digunakan adalah potongan daun ukuran 1 x 1 cm. Daun disterilisasi dengan menggunakan alkohol, bayclin dan dibilas dengan air steril. Eksplan ditumbuhkan pada media MS yang mengandung gula 30 g/l, agar 8 g/l dan vitamin group B. Penelitian dilakukan dalam 3 tahap percobaan yitu inisiasi kalus, induksi kalus embriogenik dan perkecambahan/pendewasaan kalus embriogenik. Untuk inisiasi kalus diberikan 2,4-D (0,1, 0,2 dan 0,3 mg/l).Kalus terbaik disubkultur pada media yang mengandung 2,4-D mg/l untuk mendapatkan kalus embriogenik. Kalus embriogenik yang diperoleh ditumbuhkan pada media pendewasaan dengan penambahan kinetin 91.2 dan 3 mg/l), BA (0,1:0,3 dan 0,5 mg/l) atau z eatin (0,5; 1,0 dan 1,5 mg/l) untuk induksi tunas. Parameter yang diamati adalah visual kalus yang terbentuk, persentase kalus embriogenik dan jumlah tunas. Masing-masing perlakuan disusun dengan menggunakan Rancangan Acak Lengkap dengan 10 kali ulangan. Hasil penelitian menunjukkan bahwa perlakuan 2,4-D 0,1 mg/l merupakan media terbaik untuk inisiasi kalus dengan kalus berwarna hijau keputih-putihan, agak kompak dan friabel. Subkultur kalus ke dalam media tumbuh yang sama dapat mendorong terbentuknya kalus embriogenik dengan struktur agak kompak dan berwarna putih kekuningan. Perlakuan kinetin 1 mg/l merupakan media terbaik untuk pendewasaan kalus embriogenik membentuk tunas.
